Tascam US-366 fuzz issue with DAWs

Hey guys, I'm new to the forum and have owned my Tascam US-366 for about a year now.
I've just used a microphone to record my guitar amp, and that seems to be working fine.
I've decided I want to try Cubase/Guitar Rig/Amplitube, but I'm having issues getting a good sound out of my interface.

At the moment I have:

Electric guitar - Line/Guitar.
PC speakers (3.5mm cable) - 3.5mm to 6.5mm audio converter - 2/R (1/L) out.

I'm getting terribly output from my PC speakers (crappy Logitech ones), but that shouldn't be an issue until I really crank it right? I also only have 1 cable from my PC speakers (I assume it's both mono/stereo?). I've tried plugging it into 2/R as well as 1/L - no luck with either.

Any help would gladly be appreciated. Guitar Rig and Amplitube are both giving me the same type of sound, so I think once I fix one - it will fix the other one. Any details let me know and I'll grab them.
